Abstract

This assay is a lymphocyte activation and proliferation assay that stimulates peripheral blood mononuclear cells (PBMCs) with PWM and ConA. PWM stimulates B and T lymphocytes, while ConA stimulates T lymphocytes. To assess activation of lymphocytes, the upregulation of CD69, CD40, CD80, CD86, CD40L (CD154), and CD134 is assessed. When lymphocytes are stimulated by mitogens, they undergo proliferation. Proliferating lymphoblasts are identified in flow cytometry as they appear higher on the forward and side scatter because of their increased size and complexity.
