Abstract

Zooarchaeology by Mass Spectrometry (ZooMS) is a technique that uses collagen type I and peptide mass fingerprinting to identify various taxa. Here, we share a protocol previously used with LC-MS/MS (Cleland & Vashishth, 2015; Cleland, 2018) and now adapted for MALDI-TOF and ZooMS analysis. Although it bypasses demineralization and is based on liquid chromatography principles, this protocol has yielded results comparable to acid-soluble and acid-insoluble methods when tested on bone fragments from sites with low biomolecular preservation. The protocol, called AmPhos, is named after one of its primary components, ammonium phosphate dibasic. We recommend this method for poorly preserved samples. It uses bone powder, with a suggested amount of 15-25 mg. If you use this protocol, please cite the protocol DOI, as well as the publications on which the method is based (Cleland & Vashishth, 2015; Cleland, 2018). This protocol is for research purposes only and is based on the method described in European Patent EP0323790B1.
