Abstract

The NOMADS- IR panel approach enables genomic surveillance of insecticide resistance markers in Anopheles gambiae mosquitoes using targeted nanopore sequencing. It includes amplicons targeting key insecticide resistance SNPs along targeted regions i.e. acetylcholinesterase 1(Ace1), Glutathione S-transferase epsilon 2 (Gste2), gamma-aminobutyric acid (Gaba)-gated chloride channel and voltage-gated sodium channel (Vgsc). The protocol can be done in 2 days: Day 1 - Multiplex PCR, POST-PCR DNA Clean Up, and Post-PCR DNA Quantification with Qubit steps Day 2 - End Prep and Native Sample Barcoding, Rapid Adapter Ligation and Size selection Cleanup, and Priming and loading the SpotON flow cell
