Abstract

This protocol describes a two-sample bidirectional Mendelian randomization (MR) study to test causal relationships between inflammatory skin diseases (atopic dermatitis, psoriasis, urticaria, vitiligo) and psychiatric disorders (major depressive disorder, anxiety, PTSD), comparing European (EUR) and East Asian (EAS) ancestries, with polygenic risk score (PRS) extrapolation to adolescent cohorts. Hypotheses: H1 (forward) genetic liability to inflammatory skin diseases increases depression/anxiety risk; H2 (reverse) genetic liability to depression/anxiety increases skin disease risk; H3 (cross-ancestry) causal effects differ in direction or magnitude between EUR and EAS; H4 (extrapolation) adult-derived PRS predicts adolescent depression/anxiety scales. The analysis plan follows STROBE-MR and is deposited prior to data analysis. PTSD and anxiety are analyzed in the EUR stratum only due to data availability; EAS acne is excluded due to insufficient sample size (n=1,031).
